Antonie Philips van Leeuwenhoek called the Father of Microbiology. In 1673, he observed protozoa in pond water. Microscopy is still the gold standard for parasitology. Clinical notes and travel history are essential in determining tests required. Measurements are vital for id of parasites. Ticks, mites, fleas, lice and dipteran larvae should be fixed and stored in 70 percent alcohol. Adult diptera killed with chloroform and pinned dry. To examine: mount mites directly in Hoyers solution, temporary mount for small arthropods like lice, fleas- use drop lactophenol blue or chlorlactophenol, ring with nail polish or semi-permanent mount - use melted jelly and ring with nail polish or permanent mounts- boil in 10 percent KOH or immerse in Decon 90 over night, dehydrate with alcohols (or clove oil for lice), clear methyl salicylate and mount canada balsam.
